HerpesDRG: a comprehensive resource for human herpesvirus antiviral drug resistance genotyping

The prevention and treatment of many herpesvirus-related diseases rely on antiviral therapies, but their effectiveness is often hindered by the emergence of drug resistance. Currently, the lack of a centralized database cataloging resistance mutations limits the use of sequence data for patient care. To address this gap, we developed HerpesDRG—a comprehensive drug resistance mutation database that integrates all known resistance genes and available treatment options, built from a thorough review of genotype-to-phenotype research. Alongside the Amenamevir database, we offer an R package that simplifies the annotation of resistance variants and clinical interpretation from both Sanger and next-generation sequencing data. HerpesDRG is the first open-access, community-maintainable database for drug resistance mutations in human herpesviruses (HHV), designed to support researchers and clinicians in managing HHV drug resistance.
